    摘要: 蔗糖素 (sucralose) 為一非營養性甜味劑,在全球範圍內被廣泛運用,其無熱量、高甜度的特色尤其受減肥人士及糖尿病患者的喜愛。近期研究指出蔗糖素可能對健康產生負面影響,包括影響脂質及葡萄糖代謝調控、導致氧化壓力上升等危害。粒線體在抗氧化能力調節及活性氧物種的產生均扮演重要角色,許多研究表明粒線體功能與心血管系統氧化壓力及疾病進展具密切關聯。故本研究目的為探討長期蔗糖素攝取對控制及西方飲食大鼠其心血管系統、氧化壓力及粒線體功能之影響。本研究使用48隻雄性SD大鼠,以西方飲食進行十週飲食誘導後,將控制飲食組及西方飲食組根據每日容許攝取量 (acceptable daily intake, ADI) 訂定低、高劑量 (低:30 mg/kg、高:90 mg/kg) 隨機各分為三組,每日以管餵給予蔗糖素溶液,介入十週後犧牲取其血液、心臟、主動脈樣本分析。結果顯示控制飲食組攝取10週ADI劑量之蔗糖素後,發現藉由心臟一氧化氮生合成路徑部分下調及氧化壓力的上升,導致心肌發炎現象。同時觀察到蔗糖素所引發之粒線體生合成路徑向下調節,將造成心血管系統功能受損,進而與心血管疾病風險上升具高度相關;在西方飲食組攝取10週ADI劑量之蔗糖素後,則並未觀察到顯著的心血管功能惡化情形。本研究表明蔗糖素攝取或能藉由代謝、抗氧化防禦系統及粒線體等各方面功能失調對心血管系統產生負面影響,需謹慎評估其應用。
    Sucralose, a widely used non-nutritive sweetener globally recognized for its no-calorie and intense sweetness, is particularly popular among individuals managing weight and diabetes. Recent studies have indicated potential health concerns associated with sucralose, including affecting the regulation of lipid and glucose metabolism and causing an increase in oxidative stress. Mitochondria, crucial for antioxidant capacity and reactive oxygen species (ROS) production, have been closely related to oxidative stress in cardiovascular health and disease progression. This study aimed to investigate the long-term effects of sucralose intake on cardiovascular system, oxidative stress, and mitochondrial function in Western diet-fed rats. This study used 48 male Sprague-Dawley rats, control diet and Western diet were given respectively for ten weeks of dietary induction. Then, rats from control and Western were respectively divided into low and high doses according to the acceptable daily intake (ADI) (low: 30 mg/kg, high: 90 mg/kg) via oral gavage for ten weeks. The results revealed that after ten-week sucralose intake of the control diet group at ADI levels induces cardiac inflammation through the partial downregulation of cardiac nitric oxide synthesis and an increase in oxidative stress. Additionally, it was observed that sucralose contributes to metabolic disorders and suppresses mitochondrial biogenesis, both of which adversely affect cardiovascular function and significantly elevate the risk of cardiovascular disease. However, after ten-week sucralose intake of the Western diet group at ADI levels, there was no significant deterioration in cardiovascular function. This study indicates that sucralose intake may potentially impact the cardiovascular system adversely by affecting metabolism, the antioxidant defense system and mitochondria function. Therefore, its application needs to be carefully evaluated.
