摘要: | Objective: Ulcerative colitis (UC) is an inflammatory bowel disease (IBD) characterized by continuous inflammation of the intestinal mucosa and submucosa layers. Conventional medical treatments could help to alleviate symptoms, but they can cause side effects. Therefore, exploring safer and more effective natural treatments is important. Lycium barbarum polysaccharides (LBP) can reduce colon damage, inflammation, and improve intestinal permeability. Bifidobacterium longum has been found to improve gastrointestinal symptoms and reduce inflammation. However, the effects of Bifidobacterium longum OLP 01 (OLP-01) strain on UC are rarely studied. Additionally, there is limited research on the potential synergistic effect of symbiotics combining prebiotics LBP and probiotics OLP-01 in dextran sulfate sodium (DSS)-induced UC rat models.
Methods: The duration of the experiment was 39 days. Male Sprague-Dawley (SD) rats were divided into 5 groups: normal (N), DSS-induced UC (U), UC treated with 100 mg LBP/kg bw (L), UC treated with 2 × 109 CFU OLP-01/kg bw (B), and UC treated with both 50 mg LBP/kg bw and 1×109 CFU OLP-01/kg bw (M) groups. From the first day until the end of the experiment, treatments of LBP and/or OLP-01 were administered through gavage. UC induction was conducted by administering 5% DSS continuously from day 29 to day 36, spanning a period of 7 days.
Results: LBP decreased the disease activity index (DAI) score, mitigated histological change, alleviated colon length shortening, and reduced the colonic expression of IL-6, COX-2, and iNOS with an increase in the abundance Ruminococcaceae, Lachnospiraceae, and Saccharimonadaceae families in cecum content. OLP-01 decreased the DAI score, mitigated histological change, reduced colon weight, and decreased the level of IL-17A, IL 6, COX-2, and iNOS. Enrichment of the Ruminococcus torques group genus was observed in OLP-01 treated group. Combined treatment with both LBP and OLP-01 decreased the DAI score, mitigated histological change, alleviated colon length shortening, reduced colon weight, and decreased the level of IL-17A, IL-6, and COX-2, and increased the abundance of Akkermansia, Blautia, and Ruminococcus_torques_group genera.
Conclusion: LBP and/or OLP-01 demonstrated protective effects in mitigating inflammation and modulating gut microbiota in UC, which can alleviate the progression of UC. |
