    摘要: 高尿酸血症 (hyperuricemia,HUA) 為體內累積過多尿酸 (uricacid,UA) 所造成, 不但影響腎功能,還會增加腎臟氧化壓力,其嚴重併發症可能降低病患的生活品質。 已有相關文獻證實高尿酸血症的一線用藥會對人體產生嚴重副作用,因此許多研究正 致力於探討天然化合物對預防高尿酸血症的益處。檜木醇 (hinokitiol,HIN) 是一種富 含於柏科植物的天然化合物,其抗氧化特性被認為具有治療潛力,然而,目前尚未有 檜木醇與高尿酸血症之相關研究。因此,本研究欲探討檜木醇對於高尿酸血症腎臟功 能及氧化壓力損傷之保護作用及其相關機制。預先介入大鼠檜木醇後,以氧?酸鉀 (potassium oxonate, PO) 及次黃嘌呤 (hypoxanthine, HX) 誘導高尿酸血症。犧牲時收 集血清及尿液分析生化數值及腎臟功能,並測量血清及腎臟組織的氧化壓力。為進一 步瞭解檜木醇對人類腎近曲小管細胞 (human kidney 2 cells, HK2 cells) 於高氧化壓力 下的保護效果,使用過氧化氫 (hydrogenperoxide,H2O2) 誘導高氧化壓力模式,觀察 檜木醇對腎小管細胞存活率、細胞型態及氧化壓力相關蛋白表現的影響。結果顯示, 檜木醇能顯著降低高尿酸血症大鼠的血清尿酸及肌酸酐濃度,並增加尿酸排泄分率及 肌酸酐廓清率,表示檜木醇對腎臟功能具有保護作用。此外,檜木醇有效減少體內及 腎臟氧化壓力。細胞結果也證實,檜木醇能顯著改善高氧化壓力誘導的腎小管細胞存 活率、細胞型態及氧化壓力相關蛋白表現。本研究顯示檜木醇可作為對於保護和改善 高尿酸血症一保健食品及輔助治療新選擇。
    Hyperuricemia (HUA) is caused by excess uric acid (UA) in the body, which not only affects kidney function but also increases oxidative stress on the kidneys. It may cause serious complications to patients. However, long-term use of medications for treatment may lead to severe side effects. Therefore, many studies are being conducted to explore the benefits of natural extracts in preventing HUA. Hinokitiol (HIN) is a natural compound abundant in Cupressaceae. Its antioxidant properties have therapeutic potential, but there are currently no studies on the effects of HIN on renal oxidative stress induced by HUA. This study aimed to investigate the protective effects of HIN on renal function and oxidative stress damage in HUA. After rats were pretreated with hinokitiol, the HUA model was induced with potassium oxonate (PO) and hypoxanthine (HX). Serum and urine were collected to analyze biochemical values and renal function. Measure oxidative stress in serum and kidney tissue. To further understand the protective effect of HIN on human kidney epithelial cells (HK2 cells) under high oxidative stress, hydrogen peroxide (H2O2) was used to induce high oxidative stress. The effects of HIN on HK2 cell viability, cell morphology and oxidative stress-related proteins were observed.
    The results showed that HIN can significantly reduce the serum uric acid and creatinine concentrations in hyperuricemic rats and increase the fractional excretion of uric acid (FEUA) and creatinine clearance rate (CCr), indicating that HIN has a protective effect on renal function. In addition, HIN can reduce serum and renal oxidative stress. In vitro results also confirmed that HIN can significantly improve the cell viability, cell morphology and oxidative stress-related protein expression of renal tubular cells induced by high oxidative stress. This study demonstrates that HIN can be used as a new choice for health food and auxiliary treatment to improve HUA.
