    題名: 創新保健食品VGBR抑制口腔癌生長之潛力評估
    Therapeutic Potential of Healthy Food VGBR in Alleviating Oral Cancer Progression

    摘要: 日本養生蔬菜湯(Vegetable Soup)及日本發酵玄米湯(Germinated Brown Rice)是從天然蔬果及稻米中萃取出之天然保健食品。其中含有多樣水溶性膳食纖維、維生素、植化素及GABA (γ-胺基丁酸),皆為維持人體運作、調節體質、生理機能與促進新陳代謝之不可或缺的營養素。因目前仍未有研究證實兩種天然食品萃取之保健食品是否有抑制或減緩口腔癌發生的效果,我們將上述兩種中保健食品合為一種名為VGBR之強效複合物,並在生物體外和體內(in vitro and in vivo)進行實驗,研究其抑制口腔癌的潛力。結果發現VGBR以劑量和時間依賴性方式抑制口腔癌細胞(SAS,OECM-1,HSC3)的增殖。更在傷口癒合及細胞轉移實驗中證實VGBR可以減緩口腔癌細胞的轉移能力。透過western blot實驗發現VGBR可抑制細胞週期調節因子cyclin D1和CDK4等蛋白質,間接促使癌細胞停滯於S期,並利用增強Caspase3蛋白促使癌細胞凋亡作為其抑制口腔癌細胞生長的原因。動物實驗方面,VGBR顯著降低了免疫缺陷鼠(NOD SCID)異種移植口腔癌腫瘤的生長,且沒有肝和腎毒性的不良反應。總結而言VGBR這項天然保健食品複合物可以利用多種途徑誘導細胞凋亡並抑制遷移。未來期待VGBR可以做為口腔癌患者前期的補充療法或預防補品,用簡單天然的方式抑制或預防口腔癌的進程。
    Japanese Vegetable Soup and Japanese Germinated Brown Rice are natural health supplements extracted from natural fruits, vegetables, and rice. They contain a variety of water-soluble dietary fibers, vitamins, phytochemicals, and GABA (gamma-aminobutyric acid), all of which are essential nutrients for maintaining body functions, regulating physical constitution, physiological functions, and promoting metabolism. Currently, there is no research evidence confirming whether health supplements extracted from these two natural foods have the effect of inhibiting the occurrence of oral cancer. Therefore, we combined the two health supplements into a potent compound named VGBR. We conducted experimental studies both in vitro and in vivo to investigate its potential in inhibiting oral cancer. VGBR significantly inhibits the proliferation of oral cancer cells (SAS, OECM-1, HSC3) in a dose- and time-dependent manner. Additionally, in wound healing and cell metabolism experiments, VGBR was shown to reduce the metastatic ability of oral cancer cells. Further experiments revealed that VGBR can inhibit the cell cycle regulatory factors cyclin D1 and CDK4, causing cancer cells to be arrested in the S phase. It also enhances the Caspase-3 protein, promoting apoptosis of cancer cells, contributing to the inhibition of oral cancer cell growth. In animal experiments, VGBR significantly reduced the growth of oral cancer tumors in immunodeficient mice (NOD SCID) without adverse effects of liver and kidney toxicity. In summary, VGBR, as a natural health supplement, can induce apoptosis, inhibit metabolism, and promote S phase arrest in oral cancer cells. We expected that VGBR can be used as an early-stage supplementary therapy for oral cancer patients, using simple and natural methods to inhibit the progression of oral cancer.
