    摘要: 蜂蜜屬食品且常用於中藥製劑,然而目前蜂蜜的活性成分仍不清楚,因此本研究將利用各種蜜源之蜂蜜,探討蜂蜜功效與其主要活性成分關係。利用酒精誘導胃黏膜細胞損傷試驗、促進MC3T3-E1母骨細胞礦化試驗及cisplatin誘導C2C12肌管細胞損傷試驗,作為探討蜂蜜緩中、生肌功效的實驗平台,並利用LPS誘導RAW 264.7巨噬細胞產生NO、PGE2、iNOS之發炎反應評估蜂蜜止痛作用。繼而利用抑制酪胺酸?活性試驗,探討蜂蜜的美白功效。最後配合活性組成分析結果,探討其有效成分。結果顯示蜂蜜在酪胺酸?抑制試驗中,85%以上的蜂蜜樣品(DH)對於酪胺酸?具有抑制作用,而龍眼蜜之乙酸乙酯萃取物(EAH20)抑制作用最強(IC50=2.70 mg/mL),其HPLC指紋圖譜中滯留時間為39.0分鐘為主要功效成分。乙酸乙酯蜂蜜萃取物,百花蜜(EAH17)、百花蜜(EAH18)、龍眼蜜(EAH20)可抑制巨噬細胞之NO產生,其中以EAH18功效最強(IC50=6.91 μg/mL),推測滯留時間56.5分鐘為主要功效成分。白千層蜜(EAH11)及百花蜜(EAH14)可減緩cisplatin誘導C2C12肌管細胞損傷,推測滯留時間13.5分鐘為主要功效成分。麥盧卡蜜(DH1)、百里香蜜(DH2)、百花蜜(DH3)、百花蜜(DH4)、龍眼蜜(DH5)、白千層蜜(DH11)、山瑞香蜜(DH12)、樟櫟群叢蜜(DH13)可促進骨細胞礦化,且利用乙酸乙酯萃取蜂蜜,約95%以上之萃取物可以促進骨細胞礦化,推測滯留時間19.5分鐘為主要功效成分。蜂蜜之乙酸乙酯萃取物,具有促進礦化的功能,可應用於骨頭保健之健康食品的開發,而蜂蜜則適合應用於美白化妝品。
    Honey is a food and is often used in traditional Chinese medicine preparations. However, the active ingredients of honey are still unclear. Therefore, this study will use honey from various nectar sources to explore the relationship between the efficacy of honey and its main active ingredients. Alcohol-induced gastric mucosal cell injury test, MC3T3-E1 osteocyte mineralization promotion test and cisplatin-induced C2C12 myotube cell injury test were used as experimental platforms to explore the effect of honey on invigorating spleen and muscle growth, and the analgesic effect of honey was checked on LPS-induced inflammatory model on RAW 264.7 macrophages. Then, the whitening effect of honey was explored by the experiment of inhibiting tyrosinase activity. Finally, with the analysis results of active components, discuss its active ingredients. The results show that in the tyrosinase inhibition test of honey, more than 85% of the honey samples (DH) have an inhibitory effect on tyrosinase, and the ethyl acetate extract of longan Euphoria longana Lam. honey (EAH20) has the strongest inhibitory effect (IC50=2.70 mg /mL), and its HPLC fingerprint chromatogram has a peak at 39.0 minutes as the main functional component. Ethyl acetate extracts from honey, Honey of various flowers (EAH17& EAH18), and Euphoria longana Lam. honey (EAH20) can inhibit the production of NO in macrophages, among which EAH18 has the strongest effect (IC50=6.91 μg/mL), and the peak at 56.5 minutes is the main functional ingredient. Melaleuca leucadendra honey (EAH11) and Honey of various flowers (EAH14) can slow down the injury of C2C12 myotube cells induced by cisplatin, and it is speculated that the peak at 13.5 minutes is the main functional component. Leptospermum scoparium honey (DH1), Thymus vulgaris honey (DH2), Honey of various flowers (DH3&DH4), Euphoria longana Lam. honey (DH5), Melaleuca leucadendra honey (DH11), Pittosporum tobira honey (DH12), Lauraceae honey (DH13) can promote the mineralization of bone cells. In general, more than 95% samples of EA extracts can promote the mineralization of bone cells. It is estimated that the peak at 19.5 minutes as the main functional ingredient. In conclusion, the ethyl acetate extract of honey has the function of promoting mineralization and can be used in the development of health food for bone health, while honey is suitable for whitening cosmetics.
