    摘要: 神經纖維瘤是一常見的遺傳性周圍神經系統腫瘤,目前對該疾病的治療方案有限,因此探索有效治療神經纖維瘤藥物是目標之一。近年來,透過大數據的發展加上趨近成熟的質譜技術,成為開發分離天然物策略的突破性發展,在這篇論文中,我們結合化學及生物學建立新策略,提出一個可視化的新策略,多訊息分子網絡方法 (multi-information molecular networking, MIMN),有利於更全面性、有系統的探討代謝體。本研究將針對台灣海域海綿 Lendenfeldia sp. 進行全面性的質譜分析與抗神經纖維瘤活性測試,由海綿中尋找具抗神經纖維瘤潛力之藥物。由生物試驗結果及 MIMN 闡明 Lendenfeldia sp. 海綿富含具抗炎潛力之 scalarane 骨架化合物,並由管住層析分離以及內建質譜資料庫註釋表明,所含主量化合物 24-methyl-12,24,25-trioxoscalar-16-en-22-oic acid (1) 作為主要抗趨化因子成份,主導了該海綿的抗神經纖維瘤作用。由上述結果顯示,MIMN 在探索新藥具有潛在的應用價值。
    Neurofibroma is a common hereditary peripheral nervous system tumor, and its treatment options are limited today. It is necessary to explore potential drugs for the treatment of neurofibroma. In recent years, through the development of big data and approached maturity mass spectroscopic technologies, it become the breakthrough development on new strategies of separation methods. The relevance of the study, focusing on the establishment of new strategies combining chemistry and biology, proposed a visualized method of multi-information molecular networking (MIMN), which is conducive to a more comprehensive and systematic study of metabolites. This study conducted a comprehensive metabolite analysis and anti-neurofibroma activity assessment of the Taiwanese sponge Lendenfeldia sp. The bioactive test and MINN clarified that the sponge is abundant in scalarane, which has anti-inflammatory potential. Subsequent isolation and annotation from in-house library showed that the most abundant compound, 24-methyl-12,24,25-trioxoscalar-16-en-22-oic acid (1) was used as the lead and dominated the anti-chemokine effect of sponges. The results also showed that MIMN is available in exploring new drugs.
