    Title: 膠原蛋白和維素生A酸的物化性質及其相互作用之探討

    Abstract: 膠原蛋白具有良好的保水性,膨潤性及成膜等特性,因之為製劑開發上一良好的的基
    質。膠原蛋白一般都是從小牛皮中萃取而得,而萃取的方法與過種也有所不同,本在
    萃取過種中發現ph控制在2.5 於30℃下可得適當的膨帳,並且併用酸處理及酵素消化
    豬皮時可得生體可分解性及低原性的膠原蛋白。
    為研究膠原蛋白對藥物滲透的影響,故選定維生素A 酸做為模式藥物。維生素A 酸是
    膠原蛋白代謝過程中之一中間產物,具有促進上皮細胞脫落和換新的功能,並能抑制
    角質之合成,因此近年來已被廣泛應用在皮膚外用製劑上。惟維生素A 酸對光敏感且
    難溛於水,因之本實驗利用Tween 80做為助溶劑將維生素A 酸製成水溶液製劑,並探
    討光、水量、助溶劑、氧等可能影響膠原蛋白酸安定的一系列因子。
    結果發現在30℃以下避免光的照射,可使維生素A 酸的安定性增加,另外在磷酸氫鈉
    中加入0.25% 至1%的Tween 80可使維生素A 酸的溶解度提高至一般臨床之治療劑量。
    而由滲透實驗中發現到膠原蛋白與維生素A 酸產生汲附作用,結果證實其吸咐量相當
    高,而溶液極性愈大產生滯留的現象也隨之增加。
