    摘要: 本研究利用膽固醇液晶的雙穩態特性來感測新型冠狀病毒的棘蛋白受體結 合區(SARS-CoV-2 Spike Protein Receptor Binding Domain)。此膽固醇液晶感測系 統利用蛋白質抗體抗原之免疫反應形成免疫複合體來影響膽固醇液晶的排列,並且使用偏光顯微鏡來觀察膽固醇液晶平面態與焦錐態之光學紋理變化,同時使用光譜儀與影像辨識程式來定量數據。利用微型光譜儀量測出穿透光譜,並且透過 影像辨識程式將膽固醇液晶的光學紋理數值化,以分析棘蛋白濃度與液晶紋理混 亂程度之線性關係。
    此膽固醇液晶之系統具有可以定量分析並且無需複雜的分析 儀器,可在檢測環境中進行訊號解讀與數據分析,此為新型膽固醇液晶感測系統 之特色。
    In this study, we used the bi-stable characteristic feature of cholesteric liquid crystals to detect the SARS-CoV-2 spike protein receptor-binding domain (RBD). The cholesteric liquid crystal sensing system utilizes the antibody-antigen immunoreaction to form immune complexes to affect the arrangement of cholesteric liquid crystals. Furthermore, the polarizing microscope was used to observe the optical texture changes in the planar state and the focal conic state of the cholesteric liquid crystal. Besides, the spectrophotometer and the image recognition program were utilized to quantify the concentration of the protein. In addition, the transmittance spectrum was detected through the spectrophotometer, and the optical texture of cholesteric liquid crystal was digitized via the image recognition program to analyze the linear relationship between the concentration of the spike protein and the wrinkled texture of cholesteric liquid crystal.
    In conclusion, we developed a novel sensing system using the cholesteric liquid crystal to detect the protein without the need for sophisticated analytical instrumentation, and the application can be used for the detection of spike protein in the environment soon.
