    The Application of Natural Products and their Derivatives to Preserve Islet Function Under Obesity and Pancreatic Amyloidosis Conditions

    摘要: Obesity and human islet amyloid polypeptide (hIAPP) increases the risk of β-cell dysfunction. Obesity and amyloidosis are known to be the hallmarks of type 2 diabetes (T2D). In this study, protective effects of 1,2,3,4,6-penta-O-galloyl-β-D-glucose (β-PGG) and Yakuchinone-B inhibitors were evaluated on obesity and amyloidosis in high fat diet (HFD) induced and hIAPP transgenic mice models respectively. To induce obesity by HFD, mice were provided with HFD for 12 weeks and β-PG administered with HFD. On the other hand, transgenic mice model was used for the treatment of hIAPP β-cell dysfunction in mice. Oral administration of compound 4g was carried out in transgenic mice model. In HFD mice it was observed β-PGG significantly reduced free fatty acids and Triglycerides in the adipose tissue. It was also seen that b-PGG attenuated the hypotrophy of adipose tissue in HFD. Furthermore, HFD had seen to lead hepatic steaotosis, β-PGG was seen to attenuate live damage. Additionally, β-PGG was seen to attenuate via increase in GLP-1 and adiponectin levels and also decrease in inflammatory markers such as nitric oxide synthase 2 (NOS-2). Pancreatic hypotrophy was also seen to reduce when treated with β-PGG when compared to the HFD mice group. Yakuchinune-B inhibitors were observed to have protective effects on hIAPP-induced β-cell toxicity. This further led to the use of compound 4g in transgenic mice model, treated for 6 weeks. Compound 4g reduced blood glucose levels compared to the transgenic mice group. In glycaemic parameters, the levels of glucose were observed to decrease in compound 4g treated mice. Insulin resistance which leads to T2D was seen to be ameliorated, this was further seen in a reduction in HOMA-IR and an increase HOMA-B values. β-cell hypertrophy caused by hIAPP in transgenic mice was attenuated when treated with compound 4g. In this study we concluded that β-PGG and compound 4g indicated a protective effect over HFD induced obesity and hIAPP transgenic mice.
