    摘要: Benzophenone-3 (BP-3)具有隔離紫外線的特性,主要被用於防曬乳中以及食品外包裝的油墨上,過去研究發現BP3進入環境中會干擾生物內分泌系統,是環境賀爾蒙的一種。然而,目前BP3對魚類胚胎的研究較著重於荷爾蒙及行為方面的影響,只有少數有提及魚類胚胎毒理研究。本論文分為兩個部份,首先利用斑馬魚胚胎為模式,探討BP3可能對魚類胚胎產生的毒性。胚胎浸泡於不同濃度(0、5、10、25 µM)BP3中,在96小時(4-100 hpf)急毒性試驗發現在濃度 ≥ 5 µM的暴露會造成魚鰾發育缺損、耳石縮小及側線毛細胞數目下降,25 µM的暴露會造成斑馬魚胚胎發育畸形、心臟發育功能異常、鉀離子含量降低以及皮膚細胞功能受損,以上研究結果顯示BP3暴露對魚類胚胎具有潛在的風險;第二部份研究則是利用斑馬魚成魚模式去分析BP3在不同濃度(0、11、22 µM)浸泡處理72小時後的影響,雖然在焦慮程度以及運動能力上沒有差異,但可以觀察到11、22 µM兩組處理組啄咬鏡面次數稍微增加,這顯示攻擊慾望有些微上升的趨勢。在打鬥行為實驗中11 µM組別與控制組的魚打鬥時會取得較高的勝率,之後分析取勝的原因可能是因為11 μM組別的打鬥持續時間有顯著增長因而較易取得勝率。這些結果顯示,BP3的暴露會改變魚類的社會行為,這些變化可能進一步影響水域魚類的生態環境。
    Benzophenone-3 (BP-3) has been widely used in sunscreens and many other consumer products, including cosmetics. Previous studies showed that BP3 enters the water environment and interferes the endocrine system function of fishes. However, most studies of BP3 focused on hormone function and behaviors in fish. The toxicity of BP3 in fish embryos is not fully understood. This study is divided into two parts. First, the zebrafish embryo is used as a model to explore the possible toxic effects of BP3 on fish embryos. zebrafish embryos were exposed to BP3 (0, 5, 10, or 25 µM) for 96 h (4-100 hpf), and we found that ≥ 5 µM BP3 caused developmental defects in swim bladder, otoliths, and decreased number of lateral-line hair cells. 25 µM BP3 influenced cardiac functions by decreasing stroke volume, heart rate, and cardiac output. 25 µM BP3 damaged the structure of keratinocytes in the skin of embryos. In conclusion, acute exposure of BP3 can pose adverse effects on fish embryos. The second part of this study used adult zebrafish to analyze the effects of BP3 on fish behaviors (0, 11, 22 µM). After 72 hours’ exposure, the 11 and 22 µM BP3 group slightly increased attacking behavior and the 11 µM group increased the winning rate of fighting behavior. The reasons for a higher winning rate in 11 µM group might because they have an extended fighting time. In summary, BP3 exposure might change the social behavior of fish.
