    Title: 異硫氰酸烯丙酯對於呼吸道收縮及塵蟎誘發過敏性氣喘小鼠之影響

    Abstract: 氣喘為常見的慢性呼吸道發炎性疾病,其主要病理特徵為呼吸道發炎及組織重塑,目前常見的氣喘治療藥物如皮質類固醇類藥物及β腎上腺素受體激動劑,在長期使用下會產生許多副作用。而中醫常使用三子養親湯來治療氣喘,三子養親湯含有白芥子,其經消化代謝後產生的主要代謝物為異硫氰酸烯丙酯 (allyl isothiocyanate, AITC)。因此,本次研究的目的為探討異硫氰酸烯丙酯對於塵螨 (house dust mite, HDM) 誘發之呼吸道發炎、重塑及平滑肌收縮的影響並探討其可能機制。在本離體實驗中發現,存在於芥末中的異硫氰酸烯丙酯可抑制乙烯膽鹼及鉀離子所誘發之氣管收縮。為了探討異硫氰酸烯丙酯在活體中對於過敏性小鼠的影響,本研究以鼻腔滴入塵螨萃取物的方式誘發小鼠產生過敏性氣喘,誘發方式為以連續給予五天後休息兩天為一循環進行,誘發時間總計四週,後兩週在誘發前一小時給予不同濃度之異硫氰酸烯丙酯或皮質類固醇類藥物 (dexamethasone, DEX)。相較於誘發組,介入異硫氰酸烯丙酯之組別顯著改善小鼠肺部功能、降低對塵螨特異性免疫球蛋白E (house dust mite-specific immunoglobulin E, HDM-specific IgE)、介白素4 (interleukin 4, IL-4) 與介白素5 (interleukin 5, IL-5) 含量及肺部發炎性細胞浸潤的情形。另外,實驗結果顯示異硫氰酸烯丙酯可抑制塵螨誘發增加之肌球蛋白輕鍊激酶 (myosin light-chain kinase, MLCK)、瞬態電壓感受器陽離子通道 (transient receptor potential ankyrin 1, TRPA1)、磷酸化肌球蛋白輕鍊20/肌球蛋白輕鍊20 (phospho myosin light-chain 20/myosin light-chain 20, pMLC20/MLC20)、纖連蛋白 (fibronectin)及巨噬細胞移動抑制因子 (macrophage migration inhibitory factor, MIF) 等蛋白質表現,並且可增加肺部zonula occludens-1 (ZO-1)、occludin及claudin-1表現。上述結果表明,異硫氰酸烯丙酯可能可做為治療過敏性氣喘的一個新方法。
