    Title: Study the Neuroprotective Effect of Mangosteen Pericarp Extract and Probiotic in 6-OHDA-Induced Parkinson's Disease Rat Model

    Abstract: Parkinson’s disease (PD) is the second common neurodegenerative disease in the world that impairs motor and several non-motor functions. Oxidative stress and mitochondrial dysfunction are associated with PD progression. 6-hydroxydopamine (6-OHDA) is the most frequently used neurotoxin model to study PD development as it mimics motor and several non-motor dysfunctions found in PD patients. Polyphenol rich mangosteen pericarp extract (MP) has been widely reported as potent antioxidant. Probiotics have been known to exert antioxidative properties, immunomodulator effect, and neuroprotection. Combination of probiotic and MP may increase the action of one another. This study investigated the neuroprotective effect of 8-week supplementation of low dose (LMP) and high dose (HMP) of polyphenol rich MP, and combination of LMP and probiotic Lactobacillus salivarius subsp. salicinius AP-32 (LMP+P) in 6-OHDA-induced PD rat model. Supplementation of LMP, HMP, and LMP+P prevented dopaminergic neurons loss in substantia nigra (SNc) and improved motor function in PD-like rats. This neuroprotective effect was accompanied by significant increases in the expression of antioxidant enzymes (Sod1, Sod2, Cat, GPx) and reduction of reactive oxygen species (ROS) in the brain. Further, supplementation of LMP, HMP, and LMP+P for 8 weeks prevented mitochondrial dysfunction and increased mtDNA copy number in soleus muscle. Importantly, supplementation of HMP elevated a key factor in mitochondrial biogenesis, Pgc1a, in soleus muscle. This elevation by HMP was followed by increases of mtDNA copy number and mitochondrial function in soleus muscle, and recovery of motor dysfunction. Taken together, this study shows a potential use of high dose mangosteen pericarp extract as an adjunctive therapy for PD.
