    摘要: Breast cancers can be classified into several groups depending on their expression of hormone receptors and HER2, as well as which area of the breast the tumor originates from. Triple negative breast cancer lacks expression of estrogen receptors (ER), progesterone receptors (PR) and HER2 rendering it immune to chemotherapy treatments. Consequently, patients suffering from triple negative breast cancer are subject to poor prognosis and fewer treatment options. High expression of insulin-like growth factor 1-receptor has been observed in triple negative breast cancer. IGF1R has been associated with epithelial-to-mesenchymal transition, promoting increased migration, invasion proliferation and stemness traits in breast cancer. While the link between IGF1R and breast cancer is well documented, the exact mechanisms that connect IGF1R to migration in TNBC is not yet understood. Glycosylation, one of the most common post-translational protein modifications, has the ability to alter the downstream signaling pathways of the protein being modified. There is a possibility that glycosylation of IGF1R may produce an alternative downstream signaling pathway resulting in increased migration in TNBC. The focus of this Thesis will be to not only demonstrate that IGF1R has a critical role in regulating migration in TNBC, but also to identify if glycosylation is a factor, and to ultimately identify by which mechanisms IGF1R regulates migration in TNBC. To discern a connection between glycosylation of IGF1R and migration, in addition to identifying the proteins involved in the downstream pathway towards migration may bring about new clinical approaches to treating TNBC by disabling/enabling the glycosylation of IGF1R or attacking downstream proteins that contribute to cell migration.
