    摘要: 在紅景天屬 (Rhodiola) 中共發現超過200 種物種,其中作為常用中草藥的約有20 種,例如紅景天 (Rhodiola rosea)。根據文獻,紅景天有神經保護活性 (Neuroprotection)、增強記憶與學習能力 (Learning and memory enhancement)、抗憂鬱 (Anti-depression)、抗疲勞 (Anti-fatigue)、抗發炎 (Anti-inflammatory) 以及抗氧化(Antioxidant)…等能力,而神經保護活性更是近幾年來的熱門議題。阿茲海默症 (Alzheimer’s disease) 是老年人中最常見的神經退化性疾病,且有越趨嚴重的趨勢。Methylglyoxal (MG) 在體內可造成許多的糖化反應,而糖化反應已被認為在神經退化性疾病的致病因子中扮演重要的角色,亦有許多證據顯示,糖化反應所產生的氧化壓力是阿茲海默症非常重要的致病因素。本論文建立了利用MG 誘導神經纖維瘤細胞N2A 產生退化性神經疾病的細胞模式,並評估紅景天萃取物以及從紅景天內分離得到的化合物其神經細胞保護活性。將紅景天乙醇粗萃物以水、乙酸乙酯以及正丁醇進行分配萃取,得到水層、乙酸乙酯層以及正丁醇層。對乙醯膽鹼酯酶(Acetylcholine esterase) 之抑制率分別為水層:25.8±1.2 %;乙酸乙酯層:49.2±3 %以及正丁醇層:40.8±2.9 %。MG 對N2A 所產生之細胞毒性為LC50 為3.5 mM 之濃度相關模式 (Concentrationdependentmanner),紅景天對神經細胞保護之活性分別為粗萃物:45.3±7.2 %;水層:33.8±6.0 %;乙酸乙酯層:52.8±8.8 %以及正丁醇層:51.8±8.9 %。經由乙醯膽鹼酯酶之抑制活性以及神經保護活性之篩選,乙酸乙酯層皆有最好的效果,故取乙酸乙酯層進行後續之分離純化,並搭配NMR 以及MS 進行結構鑑定,共得到15 個化合物。15 個化合物為7 個phenolic 類化合物、5 個黃酮類化合物、3個Tannin 類化合物。其中化合物RR12 (Salidroside) 在神經細胞保護活性有最好的效果,其細胞存活率為67.4±11.4 %。從實驗結果發現,紅景天為具有潛力可預防阿茲海默症之傳統中草藥。
    More than 200 species have been identified in genus Rhodiola
    (Crassulaceae). Of which about 20 species were commonly used as chinese herbal medicine such as Rhodiola rosea(R. rosea). R. rosea has been reported to have lots of activities, i.e., neuroprotective effects, learning and memory enhancement, anti-depression, anti-fatigue, antiinflammatory and antioxidant. Alzheimer’s disease(AD) is the most common type of neurodegenerative diseases in old people, and has become more severe in recent years. Methylglyoxal(MG) is able to induce glycation and play an important role in the pathogenesis of many neurodegenerative diseases. There are lots of evidences suggesting a key role of oxidative stress in the pathophysiology of AD. We use MG treated Neuro 2A cell as an in vitro model to estimate the neuroprotective effect of R. rosea. The extract from the root of R. rosea was suspended in water and partition successively with ethyl acetate (EtOAc) and n-butanol (BtOH). The inhibitory effects of aqueous layer, EtOAc layer and BtOH layer of R. rosea extract on the activity of AChE were 25.8±1.2 %, 49.2±3 % and 40.8±2.9 %, respectively. MG decreased the viability of Neuro 2A cells in a concentration-dependent manner with a LC50 of 3.5 mM and the neuroprotective effects of crude extract, aqueous layer, EtOAc layer and BtOH layer of R. rosea extract were 45.3±7.2 %, 33.8±6.0 %, 52.8±8.8 % and 51.8±8.9 %, respectively. Fifteen compounds were obtained and isolated from EtOAc layer of R. rosea. The structures were identified by NMR, MS and literature data. RR12 (Salidroside) has the best neuroprotective effect with cell viability 67.4±11.4 % of MG induced cell cytotoxicity. The results reveal that R. rosea may have a potential to prevent Alzheimer’ disease.
