    Effects of Curcuminoids on Mitochondrial Oxygen Consumption Rate and Dynamic Network in Alcohol-induced Muscle Weakness Mice

    摘要: 過量酒精攝取會引起慢性肌肉病變,症狀如肌肉無力及萎縮,會使患者出現疲勞、步伐不穩等現象,增加外傷風險。過量酒精會增加細胞氧化壓力並破壞粒線體動態平衡,使之傾向分裂,融合蛋白mitofusin (Mfn)表現量減少,嚴重時細胞可能走向內生性凋亡路徑。薑黃萃取物 (curcuminoids,CCNs)主要由curcumin、demethoxycurcumin及bisdemethoxycurcumin組成,具抗氧化功能,可防止粒線體受氧化壓力破壞,健康動物的相關實驗中發現CCNs具抗疲勞、增強抓力之效果。本研究目的為探討不同劑量之CCNs是否可減緩酒精性肌肉無力小鼠骨骼肌氧化壓力、改善粒線體功能及動態平衡並提升運動表現。實驗使用50隻雄性C57BL/6小鼠,實驗分成五組:控制組、酒精對照組、酒精及1倍CCNs組、酒精及2倍CCNs組、酒精及5倍CCNs組。結果顯示,CCNs可減緩腓腸肌重量及橫切面直徑流失、提升腓腸肌還原型態與氧化型態穀胱甘肽 (GSH/GSSG)比值。粒線體檢測結果顯示CCNs可提升耗氧速率、增加Mfn-1表現。運動表現方面可增強前肢抓力。本次實驗結果顯示,CCNs可抑制酒精引起的骨骼肌流失及氧化壓力、提升粒線體耗氧功能、增加動態平衡中融合調控蛋白之表現量並提升其運動表現。
    Ethanol abuse can cause chronic myopathy. The chronic myopathy is characterized by muscle wasting and weakness which cause fatigue, frequent falls, increasing the opportunity to get injured. Chronic ethanol consumption may increase the oxidative stress in cells, ruin the critical balance of fission and fusion of mitochondria, tend the dynamics to fission, decrease the expression of fusion-related protein, mitofusin (Mfn) and activate mitochondrial intrinsic apoptotic pathway. Curcuminoids (CCNs) comprise three major anti-oxidative phytochemicals including curcumin, demethoxycurcumin and bisdemethoxycurcumin, which possess the effect of mitochondrial protection from oxidative damage. Studies showed that curcuminoids can prevent fatigue and improve forelimb grip strength in healthy animal models. Few studies have discussed the relationship between CCNs and alcohol-induced muscle disease, especially muscle weakness. Accordingly, this study investigated the effects of curcuminoids on mitochondrial oxygen consumption rate and dynamic network in alcohol-induced muscle weakness mice. Fifty male C57BL/6 mice were randomly assigned to control group, alcohol group, alcohol plus 1X CCNs group, alcohol plus 2X CCNs group, alcohol plus 5X CCNs group. The results showed that CCNs attenuated gastrocnemius muscle weight and cross section length loss, increase serum creatinine level and GSH/GSSG ratio in gastrocnemius muscle. In mitochondrial-related tests, CCNs improved the oxygen consumption rate and up-regulated Mfn-1 expression. In physical outcomes, 5X CCNs increased the forelimb grip strength. The results suggest that 2X and 5X CCNs can attenuate alcohol-induced muscle wasting and oxidative stress, increase the mitochondrial oxygen consumption rate and fusion-related protein expression, improve the physical outcome.
