    摘要: 在各種腦神經疾病,例如中風後腦損傷或多發性硬化症、阿茲海默症、帕金森氏症、人類免疫缺乏病毒相關失憶症等神經退化性疾病中,神經膠細胞的活化及發炎反應扮演了疾病發展重要的角色。活化的神經膠細胞所分泌的各種前發炎細胞激素及神經致毒媒介會導致神經細胞的死亡,藉由抑制神經膠細胞的過度活化可以達到減緩神經退化性疾病的發展。
    Carprofen是一種非類固醇抗發炎藥物,其選擇性抑制COX-2進而降低發炎反應。本研究利用電泳酵素分析法、西方墨點法、免疫染色法等方法,發現在腦內出血,鐵的過載、細菌感染等不同腦損傷之模式下發現carprofen具有抑制MMP-9活性、COX-2與iNOS蛋白的表現。此外caprofen,也能明顯改善動物神經性評估指數、血腦障壁損傷以及腦部損傷的範圍。綜合以上結果,caprofen 可能具有應用於治療腦部發炎疾病之潛力。
    Glial cell activation plays an important role in the process of encephalopathies, such as stroke, multiple sclerosis, and Alzheimer''s disease. It has been demonstrated that the progression of neurodegenerative diseases could be slowed down by inhibiting the excessive activation of glial cells. In this study, we used NSAID drug carprofen to investigate the protective effects in different animal models of encephalopathies. Effects of intracranial injection of collagenase, LPS or Fe2+ on neurobehavior was studied in C57BL/6 mice. Post-treatment of carprofen improved sensory-motor. And the results showed that carprofen has the ability to access neurological scoring system, therefore help improve autonomous behavior, muscle strength and coordination exercise capacity results. Administration of carprofen reduced the brain MMP-9 activity in all three animal models of encephalopathy. Post-treated with carprofen could also decrease the brain inflammatory mediators (COX-2 and iNOS) in collagenase- or LPS-induced encephalopathy. However, in the Fe2+-injured model, carprofen has no significant effects on brain iNOS and COX-2 expression, although it still can improve the blood-brain barrier damage. Overall, we thought carprofen might be a drug with great potential and worthwhile to be engaged in medication development in inflammatory brain diseases.
