    摘要: 光老化是由於過度暴露在太陽光照射中,引起的彈性蛋白降解,導致皮膚彈性改變,為常見的皮膚老化現象。在本研究中,我們以臺灣產植物-MJK的莖部利用甲醇萃取並用正己烷、乙酸乙酯和正丁醇部分劃分以獲得四個分層。為了探討MJK萃取物和部分劃分之萃取物的抗老化活性,將人類皮膚纖維母細胞 (WS-1) 暴露於紫外線 (ultraviolet, UV) B之實驗模型,利用抗壞血酸作為正對照組。UVB誘導WS-1細胞光老化之能量為200 mJ/cm2,其細胞存活率在70-80%,因此利用此能量進行後續實驗。以抑制彈性蛋白酶活性和WS-1細胞內基質金屬蛋白酶 (MMP)-1活性來評估MJK萃取物之抗光老化效果。結果表明,MJK萃取物在WS-1細胞中沒有顯著的細胞毒性,且能夠保護細胞免受UVB誘導的光老化及抑制MMP-1活性。在部分劃分的萃取物中,MJK-EA和MJK-Bu抑制彈性蛋白酶活性,分別降至54.65 ± 1.41%和39.53 ± 1.57%。MJK-Bu和MJK-Aqu在WS-1細胞中沒有顯著的細胞毒性,UVB照射後細胞存活率為70.54 ± 1.45%, MJK-Bu細胞存活率達79.37 ± 3.18%,能保護細胞不受到UVB傷害,且使MMP-1活性降低至89.61 ± 5.30%。故將具抗光老化活性之MJK-Bu進行分離純化,利用Diaion HP-20管柱進行初步的分離,進一步使用Sephadex LH-20管柱和Sepbox系統的分析技術進行化合物分離。期望MJK植物可作為具抗光老化潛力之臺灣特有種植物。
    Skin aging is stimulated by elastic changes caused by exposure to solar radiation which ultimately causes photoaging. In this study, we selected the stem of Taiwanese plant-MJK, which was extracted using methanol and fractionated with n-hexane, ethyl acetate and n-butanol to obtain four fractions. In order to evaluate the anti-aging activity of MJK extract and obtained fractions an experimental model was devised, using human skin fibroblasts (WS-1) and exposing them to ultraviolet (UV) B, and ascorbic acid was used as a positive control. UVB-induced photoaging in WS-1 cells at 200 mJ/cm2 resulted in the cell viability ranging between 70-80%, hence this energy was used for all the further experiments. Anti-photoaging effect of MJK extract and its fractions was evaluated by studying the elastase activity outside the cellular system to evaluate the inhibition potential. And also by analyzing the inhibitory effect on matrix metalloproteinase (MMP)-1 in WS-1 cells. Results displayed that MJK extract exhibited no significant cytotoxicity in WS-1 cells, it was able to protect the cells from UVB-induced photoaging and was also able to inhibit MMP-1 activity. Out of all the four fractions obtained, MJK-EA and MJK-Bu inhibited elastase activity, decreasing it to 54.65 ± 1.41% and 39.53 ± 1.57%, respectively. MJK-Bu and MJK-Aqu fractions showed no significant cytotoxicity in WS-1 cells. Irradiation with UVB reduced cell viability to 70.54 ± 1.45% and MJK-Bu was able to increase the cell viability to 79.37 ± 3.18%. It was seen that MMP-1 activity was decreased to 89.61 ± 5.30% by MJK-Bu. Thus, we choose MJK-Bu for compound isolation. For isolation and purification of compounds Diaion HP-20, Sephadex LH-20 columns and the Sepbox system were used. This study implies that MJK extract and compounds can be developed into Taiwan-specific plant products with anti-photo-aging potential.
