    摘要: Abstract
    Purpose: Induction of brown-like adipocytes within white adipose tissue depots (browning) have been considered for treatment of obesity due to their ability to increase energy expenditure, and thus a wide variety of dietary agents that contribute to browning of white adipocytes are being studied. The aim of this study was to assess the effects of benzyl isothiocyanate (BITC) and phenethyl isothiocyanate (PEITC), organosulfur phytochemicals found in cruciferous vegetables, on browning of 3T3-L1 white adipocytes.
    Methods: 3T3-L1 preadipocytes were treated with different concentrations of BITC and PEITC (0.5, 1, 2, 3.5, and 5 μM) during the differentiation conditions. The lipid accumulation was determined by Oil Red O staining. The protein expression of brown-fat specific markers, including uncoupling protein 1 (UCP1), peroxisome proliferator-activated alpha (PGC1α), and PR domain containing 16 (PRDM16), were analysed by Western blot.
    Results: BITC and PEITC decreased lipid accumulation of differentiated adipocytes and significantly up-regulated protein expression of brown-adipocyte specific markers (PGC1α, UCP1 and PRDM16) in mature adipocytes undergo white adipocyte differentiation, suggesting its possible role in browning of 3T3-L1 adipocytes. Furthermore, 3T3-L1 preadipocytes treated with BITC and PEITC undergoing brown/beige adipocytes differentiation also regulation significance induction the protein expressions of brown-fat specific markers (PGC1α, UCP1 and PRDM16).
    Conclusion: BITC and PEITC can decrease lipid accumulation and increase the expressions of browning marker proteins – PGC1α, UCP1 and PRDM16. Therefore, BITC and PEITC can be valuable in the development of new drug therapies to prevent obesity with a focus on the increasing energy expenditure by fat-browning capacity.
