    摘要: 研究目的為探討枸杞多醣(Lycium barbarum polysaccharides; LBP) 與藻藍蛋白(C-phycocyanin; C-PC)對酒精誘導大鼠胃潰瘍之胃保護效果。本研究將50隻雄性Sprague-Dawley (SD)大鼠分成5組(n=10):控制組(N; normal)、負控制組(U; ulcer)、LBP組(L; ulcer+100 mg LBP /kg bw)、C-PC組(C; ulcer+50 mg C-PC /kg bw)、Mix組(M; ulcer+50 mg LBP /kg bw+25 mg C-PC /kg bw)。於第1-5週每日管餵介入物,第2-5週每兩天一次管餵50%濃度的酒精以誘導胃潰瘍,於誘導前空腹12小時,於介入後半小時進行誘導,5週後犧牲進行分析。給予LBP、C-PC與MIX可以顯著改善胃潰瘍面積,而LBP可降低血清malondialdehyde (MDA)濃度與組織IL-6、myeloperoxidase (MPO)及intracellular adhesion molecule 1 (ICAM-1)表現量。C-PC可以降低血清MDA濃度與組織tumor necrosis factor alpha (TNF-α)、IL-6、interleukin 1 beta (IL-1β)、MPO及ICAM-1表現量。Mix可降低血清MDA濃度與組織TNF-α、IL-6、IL-1β及ICAM-1表現量。LBP、C-PC和Mix也可以提升heat shock protein 70 (HSP70)與non protein sulfhydryl (NP-SH)濃度。因此,給予酒精誘導胃損傷的大鼠補充LBP、C-PC或可能藉由抗氧化、抗發炎、提升修復能力,並增加胃保護因子以達到改善胃潰瘍的效果。
    This study investigated the gastroprotective effect of Lycium barbarum polysaccharides (LBP) and C-phycoyanin (C-PC) in rats with ethanol-induced gastric ulcer . Rats were divided to 5 groups (n=10), normal (N); ulcer (U); ulcer treated with 100 mg/kg LBP (L), ulcer treated with 50mg/kg C-PC (C); and ulcer treated with mix of 50 mg/kg LBP and 25 mg/kg C-PC (MIX; M). Pretreatment were given from 1st to 5th week, and ulcer induction were performed every other day from week 2nd to 5th week with 50% ethanol level. Rats were sacrificed after last induction; gross evaluation of gastric tissue, analysis of serum and gastric tissue were performed. LBP or C-PC attenuated the severity of gastric mucosal damage was observed in histopathology. LBP decreased serum MDA levels and tissue IL-6, MPO, and ICAM-1 level. C-PC decreased serum MDA levels and tissue TNF-α, IL-6, IL-β, MPO and ICAM-1 levels. MIX decreased serum MDA levels and tissue TNF-α, IL-6, IL-1β and ICAM-1 level. LBP, C-PC or MIX increased gastric HSP 70 and NP-SH levels. Our study results suggested that pretreatment of LBP and C-PC reduced ethanol-induced gastric injury in rats through suppression of gastric inflammation, oxidative stress and elevation of gastroprotective substance.
