    摘要: 本篇研究目的為探討高不飽和脂肪酸與飽和脂肪酸比值 (P/S ratio) 搭配不同含量的單元不飽和脂肪酸對3T3-L1脂肪細胞脂質生合成及脂質相關代謝之作用機轉。3T3-L1前脂肪細胞先培養於含dexamethasone、3-isobutyl-1-methylxanthine及insulin的Dulbecco’s modified eagle medium (DMEM) 培養液中,再以含insulin的培養液培養使其分化為成熟脂肪細胞後,分別用體積比1: 10: 5及1: 2.5: 5的方式混合棕櫚酸、油酸及亞麻油酸培養脂肪細胞 (相等於P/S ratio=5; MUFA 60%,簡稱M60及P/S ratio=5; MUFA 30%,簡稱 M30) 。經過72小時後,以油紅染色分析脂肪酸對細胞內脂質堆積程度的影響,並分析細胞內三酸甘油酯 (TG) 的含量; 同時分析培養基中甘油的釋出量以及利用real-time PCR的方法分析脂質代謝相關基因peroxisomal proliferator-activated receptor γ (PPAR γ) 、leptin、adiponectin、glucose transporter-4 (GLUT-4) 、hormone-sensitive lipase (HSL) 及lipoprotein lipase (LPL) 的表現。結果發現,10 μM低劑量的M60組 (M60-10) 與同劑量的M30組 (M30-10) 相比,可顯著降低細胞內TG含量以及PPAR γ、LPL、leptin、adiponectin、GLUT-4 mRNA的表現量,同時能增加脂解作用相關酵素HSL mRNA的表現,進而達到降低脂肪細胞三酸甘油脂蓄積的效果。
    The development of obesity is characterized by an increase in the number of fat cells and their lipid content, as a result of adipogenesis and differentiation. In this study, we determined the effects of high polyunsaturated to saturated fatty acid ratio (P/S ratio=5) and different amount of monounsaturated fatty acid (MUFA=30% or 60%) on adipogenesis in 3T3-L1 adipocytes. 3T3-L1 preadipocytes were cultured in differentiated medium for 12 days to differentiate into mature adipocytes. Different ratio of palmitic acid (PA), oleic acid (OA) and linoleic acid (LA) will first bound with bovine serum albumin (BSA) before given to 3T3-L1 adipocytes in low dose and high dose. Results showed that low dose of P/S ratio=5, MUFA=60% (M60) fatty acid could decrease the accumulation of triglycerides in mature adipocytes by decreasing the mRNA expression of adipogenic factors such as peroxisome proliferator-activated receptors (PPARs) , lipoprotein lipase (LPL) and glucose transporter-4 (GLUT-4) whereas the lipolytic enzymes (hormone sensitive lipase, HSL) expression was increased. Furthermore, the treatment of M60 in low dose also decreased the secretion of leptin and increased the secretion of adiponectin in adipocytes. In summary, the composition of P/S=5, MUFA 60% fatty acid in low dose seems to have anti-adipogenic effects on 3T3-L1 adipocytes due to the down-regulation of adipogenic effects and transcription factors.
