    摘要: 近年來,根據許多國家的流行病學研究指出肥胖與多種癌症的發生率有相關,如大腸直腸癌、胃癌、食道癌、肝癌、膽囊癌、胰臟癌、腎臟癌、子宮癌,以及女性乳癌等。目前已有許多研究顯示脂肪組織中的脂肪間質幹細胞,其生物活性對癌症具有一定的影響。然而目前脂肪間質幹细胞對於腫瘤進程的確切的作用仍有爭議。因此,我們的實驗目標是研究脂肪間質幹細胞使否透過分泌細胞激素抑或直接與不同惡性腫瘤細胞之間的產生相互作用,如促進腫瘤生長。另外,癌初始細胞被認為是癌症接受放射治療和化學治療後,發生癌症復發的原因,所以我們也將針對脂肪間質幹細胞是否會促進癌初始細胞的特性進行探討,透過細胞球體培養的實驗評估癌初始細胞的特性,發現脂肪間質幹細胞能增強球體形成生長。最後在基因和蛋白質分析顯示,脂肪間質幹細胞會通過第六型介白素活化促進癌細胞的惡性發展。
    Recently, obesity has been demonstrated that is correlated with the progression of various cancer types, such as colorectal cancer, stomach cancer, esophagus cancer, liver cancer, gallbladder cancer, pancreas cancer, kidney cancer, uterine cancer and breast cancer, according to epidemiological research in many countries. The biological activity of mesenchymal stem cell in adipose tissue has been shown to influence tumor development. However, the exact effects of adipose-derived mesenchymal stem cells on tumor progression remain controversial. Thus, the aim of this study is to investigate whether adipose-derived mesenchymal stem cells affect tumor development through direct interaction or cytokine secretion. Moreover, the recurrence of cancer after radio- or chemo-therapy has been considered that is result from cancer-Initiating cells. We also investigated the facilitating role of adipose-derived mesenchymal stem cells toward cancer-Initiating cells in this study. We evaluate the characteristics of cancer-Initiating cells by sphere formation assay. The result of sphere formation assay showed that adipose-derived mesenchymal stem cells can enhance the formation of tumor sphere. In the end, the results of gene and protein expression showed that adipose-derived mesenchymal stem cells can promote the malignancy of cancer cell through interleukin 6-related pathway.
