    Title: 量子點CeTe和CdTe/CdS對於人類大腸癌細胞的毒性測試

    Abstract: 量子點是一種奈米粒子,藉由半導體金屬合成出來,具有嶄新的物化特性。例如:高效能的螢光量子產率(photoluminescence quantum yield),顯著的史托克位移(Stokes shift),範圍廣的吸收光譜且具有對稱並狹窄的螢光光譜(photoluminescence spectra, PL spectra),光穩定性佳,可用單一激發波長,其螢光隨著粒徑大小不同呈現不同螢光顏色。
    本研究主要探討量子點QDs(CdTe與CdTe/CdS)對於人類大腸癌細胞株(WiDr)是否具有引發細胞死亡之作用、造成死亡的相關機制及量子點進入細胞後是否會聚集於特定位置。因此,利用MTT assay測定細胞粒線體代謝活性以及流式細胞儀分析技術了解細胞凋亡的情形,同時也觀察細胞產生含氧自由基(ROS)的含量變化,由實驗中得知,隨著加入的量子點QDs濃度增加,造成細胞生存率下降,而細胞凋亡(apoptosis)與含氧自由基(ROS)的程度則逐漸增加。接著使用雷射共軛焦顯微鏡(confocal microscope),觀察量子點進入到細胞後位於細胞質,最後,利用飛行式二次離子質譜儀(ToF-SIMS)來觀察當人類大腸癌細胞株(WiDr)處理量子點QDs(CdTe與CdTe/CdS)後,Cd和Te在細胞內的分布。藉由本研究可以提供量子點QDs(CdTe與CdTe/CdS)對於in vitro細胞實驗造成的細胞毒性測試有相當的了解。
