    題名: 山藥儲藏性蛋白質與其合成胜肽對於高脂飼料誘導肥胖小鼠的研究

    摘要: 根據衛生福利部102年統計資料顯示,腦血管疾病、心臟疾病、糖尿病、高血壓性疾病及腎病變皆位居國人十大死因之列。而這些歸類為代謝症候群之慢性疾病與肥胖有關,儼然成為台灣相當重要且急須面對之健康議題。因此如能有效預防與降低肥胖症之發生,對人民健康即能有所助益。本研究室先前已經透過各項實驗指出山藥儲藏性蛋白具有調節血壓活性、清除自由基能力以及改善葡萄糖不耐受性。本研究將繼續探討山藥儲藏性蛋白抑制脂肪堆積能力。以高脂肪飼料餵食C57BL/6 小鼠,同時間餵食山藥儲藏性蛋白80 mg/Kg及其合成胜肽NW 40 mg/Kg,共138天。結果顯示在第14天後高脂肪飼料配合山藥儲藏性蛋白及其胜肽兩組明顯減緩體重的增加並與高脂飼料組呈現顯著差異(P <0.05)。分析進食量,高脂肪飼料組(三組)皆無顯著差異;飼料轉換率在高脂肪飼料配合給予山藥儲藏性蛋白及其合成胜肽NW之兩組卻高於單純攝食高脂肪飼料組(即兩組相對需攝食較高量的高脂肪飼料才能增加1g重量);分析葡萄糖耐受性,山藥儲藏性蛋白或合成胜肽NW配合高脂飼料顯著改善葡萄糖不耐性;分析血液生化指標參數,高脂肪飼料配合給予山藥儲藏性蛋白及其合成胜肽NW兩組各自降低了血液中總膽固醇含量以及三酸甘油脂含量。最後從基因表現量的情況來看,山藥儲藏性蛋白與其合成胜肽NW皆能夠抑制FAS基因的表現進而減少脂肪的形成。

    The Ministry of Health and Welfare reported that the metabolic disease,including cerebrovascular disease, heart disease, diabetes, nephropathy and hypertension, are ranked among the top ten death causes in Taiwan.Which these chronic diseases are closely associated with obesity. Therefore,obesity has become a very important health issue in Taiwan and the urgent need to confront it.
    We have previously confirmed that yam storage protein of dioscorin has been proved to have the activity of regulating blood pressure, free radical scavenging capacity and improvement of impaired glucose tolerance. The propose of this study is to investigate the inhibition of fat accumulation by yam storage protein of dioscorin and synthesized dipeptide NW in diet-inded obese mice.The oral administration of dioscorin (80 mg/Kg) or synthesized dipeptide NW (40 mg/Kg) daily concurrent with high-fat (HF) diets for 138 days.The present results show that the body weight of 2-week dioscorin or synthesized dipeptide NW interventions were showed significantly lower (P < 0.05) compared to the HF group.There were no significant differences between HF group and (HF+ dioscorin) or (HF+ NW) of food intake but the feed conversion rate of the dioscorin and synthized dipeptide NW were higher than HF group.It means that this two group need more HF diet to gain the weight. The dioscorin intervention (HF+ dioscorin) or synthesized dipeptide NW intervention (HF+NW) was showed significantly different (P < 0.05) and improved the impaired glucose tolerances compared to HF group in obese mice by the oral glucose tolerance tests. It was also found that dioscorin interventions lower the total cholesterol and dipeptide NW lower the triglyceride compared to the HF group of the blood biochemical index. The dioscorin and synthesized dipeptide NW can reduced the fat by inhibiting the FAS gene expression.
