    Title: 牛樟芝與扁桃斑鳩菊活性成分探討
    Study on the Constituent of Antrodia camphorata and Vernonia amygdalina Delile

    Abstract: 本研究主要探討多孔菌科薄孔菌屬牛樟芝子實體及菊科斑鳩菊屬扁桃斑鳩菊莖部之化學成分,以了解植物中成分與活性之間的關連性。將牛樟芝酒精萃取物直接進行管柱層析;將扁桃斑鳩菊萃取物溶於水,且再以n-hexane、EtOAc、n-butanol分別由低極性到高極性進行分配萃取,得到各劃分層再經過不同溶煤系統以管柱層析分離純化後,總共分離純化出三十四個化合物。從牛樟芝95%EtOH / H2O之萃取物得到十七個化合物為2-hydroxy, 5-hydroxymethyl furan (1) 2-formyl, 5-hydroxy methyl furan (2)、4-hydroxy phenyl acetate (3)、2,3-(methylenedioxy)-4-methoxy-5-methyl phenol (4)、4, 7-dimethox-5-methyl-1, 3-bezodioxole (5)、2,3-dimethyl-5-methyl-[1,4]-benzoquinone (6)、4-hydroxysesamin (7)、Zhankuic acid A (8)、Zhankuic acid B (9)、Zhankuic acid C (10)、Antcin C (11)、Antcin K (12)、Ergosterol (13)、Dehydroeburicoic acid (14)、Dehydrosulphurenic acid (15)、15α-acetyl-dehydrosulphurenic acid (16)、CoQ3 (17)。另外,扁桃斑鳩菊莖部MeOH之萃取物,於乙酸乙酯層中分離純化出的化合物為(Z)-2-hydroxy benzyl) butenedioic acid (19)、methyl caffeate (18)、3, 5-di-O-caffeoyl-quinic acid (23)、methyl 3, 4-di-O-caffeoyl-quinic acid (24)、3, 4, 5-tri-O-caffeoyl-quinic acid (26)、luteolin (27)、chrysoeriol-7-O-B-D-glucopyranoside (28)、VA-1 (30);另由正丁醇層分離出(Z)-2-hydroxy benzyl) butenedioic acid (19)、chlorogenic acid (20)、1, 3-di-O-caffeoyl-quinic acid (21)、3, 4-di-O-caffeoyl-quinic acid (22)、3, 5-di-O-caffeoyl-quinic acid (23)、methyl 3, 5-di-O-caffeoyl-quinic acid (25)、syringaresinol-4-O-B-D-glucopyranoside (29)、VA-1 (30)、Vernonioside B4 (31)、VE-1 (32)、Vernonioside A5 (33)、Vernonioside E2 (34),其中化合物30、31、33、34為新化合物。本次分離所得化合物進一步進行L6細胞對葡萄糖攝入活性試驗,實驗結果投予化合物1、2、5、6、8、17、21、31效果優於insulin;另外,同時進行對金黃色葡萄球菌抑菌試驗,結果顯示加入化合物16、24、26、27有良好的抑菌效果。

    In this study, constituents of wild type and cultivated fruting body of Antrodia camphorata and stem of Vernonia amygdalina Delile were investigated. The ethanol extract of A. camphorata chromatograph directly. The extract of V. amygdalina dissolved in water and partitioned with n-hexane、EtOAc、n-butanol from lower to higher polarity. Total compounds were isolated with different kind of chromatography and solvents system. Total of 34 compounds were isolated. There are 17 compounds isolated from A. camphorata:2-hydroxy, 5-hydroxymethyl furan (1)、2-formyl, 5-hydroxymethyl furan (2)、4-hydroxy phenyl acetate (3)、2,3-(methylenedioxy)-4-
    methoxy-5-methyl phenol (4)、2,3-(methylenedioxy)-1, 4-dimethoxy-6-
    methyl phenol (5)、2,3-dimethyl-1,4-benzoquinone (6)、4-hydroxysesamin (7)、Zhankuic acid A (8)、Zhankuic acid B (9)、Zhankuic acid C (10)、Antcin C (11)、Antcin K (12)、Ergosterol (13)、Dehydroeburicoic acid (14)、Dehydrosulphurenic acid (15)、15α-acetyl-dehydrosulphurenic acid (16)、CoQ3 (17)。The stem of Vernonia amygdalina extracted by MeOH. In EA layer, we got 8 compounds:(Z)-2-hydroxybenzyl) butenedioic acid (19)、methyl caffeate (18)、3, 5-di-O-caffeoyl-quinic acid (23)、methyl 3, 4-di-O-caffeoyl-quinic acid (24)、3, 4, 5-tri-O-caffeoyl-quinic acid (26)、luteolin (27)、chrysoeriol-7-O-B-D-glucopyranoside (28)、VA-1 (30);in the butanol layer: (Z)-2-hydroxy benzyl) butenedioic acid (19)、chlorogenic acid (20)、1, 3-di-O-caffeoyl-quinic acid (21)、3, 4-di-O-caffeoyl-quinic acid (22)、3, 5-di-O-caffeoyl-quinic acid (23)、methyl 3, 5-di-O-caffeoyl-quinic acid (25)、syringaresinol-4-B-D-glucopyranoside (29)、VA-1 (30)、Vernonioside B4 (31)、 VE-1 (32)、Vernonioside A5 (33)、Vernonioside E2 (34),and 30、31、33、34, those of them are new compounds。The compound what we separated from this experiment is going to glucose uptake via L6 cell line bioassay. Giving compounds 1、2、5、6、8、17、21、31 shows better effect than insulin in the result;In addition, going to test the inhibition of Straphylococcus aureus at the sametime. The result shows that adding compound 16、24、26、27 has good effect of bacteria inhibition.
