    Title: β—胡蘿蔔素對DMBA誘發雌鼠乳腺腫瘤生成之延緩效應

    Abstract: 本研究係探討β-胡蘿蔔素對DMBA誘發乳腺腫瘤之延緩效應。以Sprague-Dawley雌鼠為實驗動物模式,隨機分為四組,分別為餵食AIN-93標準飲食之對照組、以及額外添加3種不同劑量之β-胡蘿蔔素的實驗組,分別為0.1%、0.4%、0.6% β-胡蘿蔔素組。於鼠齡7週大時以相當老鼠每公斤體重40毫克之7,12-dimethylbenz(a)anthrancene (DMBA)灌食來誘發其乳腺腫瘤之生成,並在誘導後2週開實驗組老鼠始餵食前述之不同劑量之飲食並記錄攝食量。至鼠齡90天起,每週進行觸診,記錄腫瘤開始出現的時間、大小、位置、及生長狀況。於DMBA誘導26週後犧牲,分析血漿及肝臟中之β-胡蘿蔔素、維生素A及維生素E濃度,和紅血球及肝臟中抗氧化酵素超氧歧化酶(superoxide dismutase, SOD)、過氧化氫酶(catalase, CAT)、麩胱甘肽還原酶(glutathione reductase, GRd)、麩胱甘肽過氧化(glutatione peroxidase, GPx)及抗氧化物質麩胱甘肽(glutathione, GSH)濃度,並取乳腺腫瘤秤重做組織切片分析,另外亦取出肝、肺、腎、卵巢做切片檢查,觀察是否有腫瘤轉移的現象。結果發現飲食中額外添加β-胡蘿蔔素,對乳腺腫瘤之大小及重量沒有影響,但可增加腫瘤發生之潛伏期使腫瘤發生得以延緩,而體內SOD和GPx活性的提昇及GSH濃度的增加,推測其機制可能和其體內抗氧化能力提昇所致。而使用之劑量,以添加0.6% β-胡蘿蔔素組之效果最佳。
